Turpinal® products are high purity grades of etidronic acid and sodium etidronate which are suitable for use in cosmetics or any applications requiring a very low metal content.

Turpinal SL high purity of Turpinal® is guaranteed by thorough quality control procedures (including testing for microbiological contamination). 
Moreover, the certificate of analysis accompanying each batch contains detailed information covering chemical purity and trace metal content.

Turpinal® SL and Turpinal® 4NL are respectively the acid form and the tetra sodium salt of etidronic acid. 
They are colorless to slightly yellow liquids with a neutral odor. 
Turpinal® 4NP is the tetrasodium powder form of etidronic acid.

These products can be mixed with water in all ratios and have a strong complexing capacity in relation to polyvalent cations. 
The excellent complexing capacity for metals is exploited for the stabilization of H2O2-containing preparations and other preparations with active oxygen. 
The product is also suited for application in mercaptan – containing formulations and enhances the color stability of bar soaps. 
Turpinal SL is recommended that Turpinal® be added during production with a concentration of 0.1 – 2% with reference to the finished preparation.

TURPINAL® GENERAL FEATURES:

Properties, applications: 
Complexing properties make Turpinal products a remarkable candidate for complexing metal ions, especially in transition metals (Fe, Cu, Mn, Zn) and water hardness ions (Ca, Mg). 
Turpinal's high complex strength allows to limit or even prevent precipitation of iron hydroxide. 
Copper also forms a very strong complex with Turpinal, and the formation and precipitation of copper hydroxide can be remarkably reduced and sometimes completely avoided. 
Turpinal SL is not always necessary to operate at stoichiometric complexing concentrations to prevent calcium scaling and precipitation. 
In fact, in many cases, calcium crust formation can be inhibited by sub-stoichiometric amounts of Turpinal. 
This feature is known as the 'threshold effect'.
Due to these complexing properties and the threshold effect, as well as its chemical stability in the oxidizing environment, Turpinal can show material benefits in many fields of application, more particularly in the cosmetic industry.

Toxicological profile: Turpinal showed low toxicity in both acute and chronic tests. 
No mutagenic or sensitization properties were found.
Storage and transportation: Turpinal 4NL and Turpinal SL have a shelf life of at least 1 year in their original sealed packaging at 5-30°C. 
Only containers made of V4A steel and polyethylene (taking into account the corrosive effect) are suitable for transport and storage. 
Turpinal 4NL and Turpinal SL are not sensitive to temperature changes. 
However, these solutions should be kept from cold to prevent crystallization of the product.

Turpinal SL - General Information
A diphosphonate which affects calcium metabolism. It inhibits ectopic calcification and slows down bone resorption and bone turnover. [PubChem]

Pharmacology of Turpinal SL
Turpinal SL is a first generation (non-nitrogenous) bisphosphonate in the same family as clodronate and tiludronate. 
Turpinal SL affects calcium metabolism and inhibits bone resorption and soft tissue calcification. 
Of the etidronic acid that is resorbed (from oral preparation) or infused (for intravenous drugs), about 50% is excreted unchanged by the kidney. 
The remainder has a very high affinity for bone tissue, and is rapidly absorbed onto the bone surface. 
Turpinal SL has been shown to prevent or delay skeletal-related events and decrease bone pain as well as normalize calcium levels in the presence of hypercalcemia.

There have been isolated reports of patients experiencing increases in their prothrombin times when etidronate was added to warfarin therapy. 
The majority of these reports concerned variable elevations in prothrombin times without clinically significant sequelae. 
Although the relevance of these reports and any mechanism of coagulation alterations is unclear, patients on warfarin should have their prothrombin time monitored.

Turpinal SL Contraindications
Didronel tablets are contraindicated in patients with known hypersensitivity to etidronate disodium or in patients with clinically overt osteomalacia.

Turpinal SL Indication: For the treatment of symptomatic Paget's disease of bone and in the prevention and treatment of heterotopic ossification following total hip replacement or due to spinal cord injury.
Mechanism Of Action: Bisphosphonates, when attached to bone tissue, are absorbed by osteoclasts, the bone cells that breaks down bone tissue. 
Although the mechanism of action of non-nitrogenous bisphosphonates has not been fully elucidated, available data suggest that they bind strongly to hydroxyapatite crystals in the bone matrix, preferentially at the sites of increased bone turnover and inhibit the formation and dissolution of the crystals. 
Other actions may include direct inhibition of mature osteoclast function, promotion of osteoclast apoptosis, and interference with osteoblast-mediated osteoclast activation. 
Turpinal SL does not interfere with bone mineralization. 
In malignancy-related hypercalcemia, etidronic acid decreases serum calcium by inhibiting tumour-induced bone resorption and reducing calcium flow from the resorbing bone into the blood. 
Turpinal SL also reduces morbidity of osteolytic bone metastases by inhibiting tumour-induced bone resorption. 
Turpinal SL may promote osteoclast apoptosis by competing with adenosine triphosphate (ATP) in the cellular energy metabolism. 
The osteoclast initiates apoptosis and dies, leading to an overall decrease in the breakdown of bone.

Absorption: The amount of drug absorbed after an oral dose is approximately 3%.
Toxicity (Overdose): Clinical experience with acute etidronic acid overdosage is extremely limited. 
Decreases in serum calcium following substantial overdosage may be expected in some patients. 
Signs and symptoms of hypocalcemia also may occur in some of these patients. 
Some patients may develop vomiting. In one event, an 18-year-old female who ingested an estimated single dose of 4800 to 6000 mg (67 to 100 mg/kg) of etidronate was reported to be mildly hypocalcemic (7 .5 2 mg/ dl) and experienced paresthesia of the fingers.

For relief of symptoms of dysuria, urgency, nocturia, suprapubic pain, frequency and incontinence as may occur in cystitis, prostatitis, urethritis, urethrocystitis/urethrotrigonitis.
Turpinal SL Pharmacology
Flavoxate is a spasmolytic flavone derivative that acts by relaxing the smooth muscle of the urinary tract. 
Flavoxate is a competitive muscarinic receptor antagonist indicated for the treatment of overactive bladder with symptoms of urge urinary incontinence, urgency and urinary frequency. 
Muscarinic receptors play important roles in several major cholin-ergically mediated functions, including bladder smooth muscle contractions and stimulation of salivary secretion.

Turpinal SL Toxicity
The oral LD50 for flavoxate HCl in rats is 4273 mg/kg. The oral LD50 for flavoxate HCl in mice is 1837 mg/kg. 
Overdose symptoms include seizures, decreased ability to sweat, (hot, red skin, and dry mouth, and a high body temperature), hallucinations, increased heart rate and blood pressure, and mental confusion.

WHAT IS TURPINAL SL?
Turpinal SL (Turpinal SL) belongs to a group of medicines called bisphosphonates (bis FOS fo nayts). 
It alters the cycle of bone formation and breakdown in the body.
Turpinal SL is used to treat Paget's disease, and to treat conditions of irregular bone growth due to hip fracture or spinal cord injury.
Turpinal SL may also be used for purposes not listed in this medication guide.

Turpinal SL disodium tablets, USP are indicated for the treatment of symptomatic Paget’s disease of bone and in the prevention and treatment of heterotopic ossification following total hip replacement or due to spinal cord injury. 
Turpinal SL disodium tablets are not approved for the treatment of osteoporosis.

Paget’s Disease
Turpinal SL disodium tablets are indicated for the treatment of symptomatic Paget’s disease of bone. 
Turpinal SL disodium therapy usually arrests or significantly impedes the disease process as evidenced by:

•Symptomatic relief, including decreased pain and/or increased mobility (experienced by 3 out of 5 patients).
•Reductions in serum alkaline phosphatase and urinary hydroxyproline levels (30% or more in 4 out of 5 patients).
•Histomorphometry showing reduced numbers of osteoclasts and osteoblasts, and more lamellar bone formation.
•Bone scans showing reduced radionuclide uptake at pagetic lesions.
In addition, reductions in pagetically elevated cardiac output and skin temperature have been observed in some patients.

In many patients, the disease process will be suppressed for a period of at least one year following cessation of therapy. 
The upper limit of this period has not been determined.

The effects of the Turpinal SL disodium treatment in patients with asymptomatic Paget’s disease have not been studied. 
However, Turpinal SL disodium treatment of such patients may be warranted if extensive involvement threatens irreversible neurologic damage, major joints, or major weight-bearing bones.

Heterotopic Ossification
Turpinal SL disodium tablets are indicated in the prevention and treatment of heterotopic ossification following total hip replacement or due to spinal cord injury.

Turpinal SL disodium tablets reduce the incidence of clinically important heterotopic bone by about two-thirds.
Among those patients who form heterotopic bone, Turpinal SL disodium tablets retard the progression of immature lesions and reduces the severity by at least half.

In total hip replacement patients, Turpinal SL disodium tablets do not promote loosening of the prosthesis or impede trochanteric reattachment.
In spinal cord injury patients, Turpinal SL disodium tablets do not inhibit fracture healing or stabilization of the spine.
Turpinal SL is used to treat Paget’s disease (a condition in which the bones are soft, weak and may be deformed, or easily broken), Hypercalcemia of malignancy (abnormally high calcium levels in blood resulting from cancer), Heterotopic Ossification (presence of bone in soft tissue where it does not exist normally).

HOW SHOULD I USE TURPINAL SL?
Use Turpinal SL as directed by your doctor. Check the label on the medicine for exact dosing instructions.

Take Turpinal SL with 6 to 8 ounces (180 to 240 mL) of plain water first thing in the morning, at least 30 minutes before eating, drinking, or taking any other medicine. 
Turpinal SL works best if taken 60 minutes before eating, drinking, or taking any other medicine. 
Taking Turpinal SL with orange juice, coffee, or mineral water may decrease its effectiveness.
Swallow whole. Do not break, crush, suck, or chew the tablet before swallowing.
Do not lie down for at least 30 minutes after taking Turpinal SL and until after you have had your first meal or snack of the day.
Do not take Turpinal SL at bedtime or before getting out of bed.
Calcium or iron supplements, vitamins, or antacids containing calcium, magnesium, or aluminum may interfere with the absorption of Turpinal SL. 
These medicines must be taken at least 2 hours after taking Turpinal SL.
If you miss a dose of Turpinal SL, skip the missed dose and go back to your regular dosing schedule. 
Do not take 2 doses at once.

Etidronic acid, also known as etidronate, is a bisphosphonate used as a medication, detergent, water treatment, and cosmetic.

Turpinal SL was patented in 1966 and approved for medical use in 1977.

Use
Medical
Etidronic acid is a bisphosphonate used to strengthen bone, treat osteoporosis, and treat Paget's disease of bone.

Bisphosphonates primarily reduce osteoclastic activity, which prevents bone resorption, and thus moves the bone resorption/formation equilibrium toward the formation side and hence makes bone stronger on the long run. 
Etidronate, unlike other bisphosphonates, also prevents bone calcification. 
For this reason, other bisphosphonates, such as alendronate, are preferred when fighting osteoporosis. 
To prevent bone resorption without affecting too much bone calcification, etidronate must be administered only for a short time once in a while, for example for two weeks every 3 months. 
When given on a continuous basis, say every day, etidronate will altogether prevent bone calcification. 
This effect may be useful and etidronate is in fact used this way to fight heterotopic ossification. 
But in the long run, if used on a continuous basis, it will cause osteomalacia.

Chemical
Main article: Corrosion inhibitor
HEDP is used as a retardant in concrete, scale and corrosion inhibition in circulating cool water system, oil field and low-pressure boilers in fields such as electric power, chemical industry, metallurgy, fertilizer, etc. In light woven industry, HEDP is used as detergent for metal and nonmetal. 
In dyeing industry, HEDP is used as peroxide stabilizer and dye-fixing agent; In non-cyanide electroplating, HEDP is used as chelating agent. 
Turpinal SL dosage of 1–10 mg/L is preferred as scale inhibitor, 10–50 mg/L as corrosion inhibitor, and 1000–2000 mg/L as detergent. 
Usually, HEDP is also used together with polycarboxylic acid (superplasticizer), in which it acts as reducing agent.

Etidronic acid is a chelating agent and may be added to bind or, to some extent, counter the effects of substances, such as calcium, iron or other metal ions, which may be discharged as a component of grey wastewater and could conceivably contaminate groundwater supplies. 
As a phosphonate it has corrosion inhibiting properties on unalloyed steel.
Etidronic acid also acts to retard rancidification and oxidation of fatty acids.

HEDP and its salts are added to detergents and other cleaning agents to prevent the effects of hard water. 
Turpinal SL is also used in peroxide bleaching to prevent degradation of peroxides by transition metals.

Etidronic acid is listed as an ingredient of several cosmetic formulations where it is used for suppressing radical formation, emulsion stabiliser and viscosity control. 
While etidronic acid has not been limited from inclusion in cosmetics and does have legitimate uses, it is recommended that, as with most cosmetic products (particularly soaps), the product should be thoroughly rinsed from the skin after use.

Etidronic acid is also included among swimming pool chemicals. 
Turpinal SL is used as a stain inhibitor to prevent metal ions coming out of solution and staining the sides of swimming pools.

9 Usage:
HEDP is used as scale and corrosion inhibition in circulating cool water system, oil field and low-pressure boilers in fields such as electric power, chemical industry, metallurgy, fertilizer, etc.. 
Turpinal SL light woven industry, HEDP is used as detergent for metal and nonmetal. 
Turpinal SL dyeing industry, HEDP is used as peroxide stabilizer and dye-fixing agent; In non-cyanide electroplating, HEDP is used as chelating agent. 
Turpinal SL dosage of 1-10mg/L is preferred as scale inhibitor, 10-50mg/L as corrosion inhibitor, and 1000-2000mg/L as detergent. Usually, HEDP is used together with polycarboxylic acid.
