GLYCERYL ESTERS

Glyceryl esters, also known as glycerides, are a class of esters formed from glycerol (propane-1,2,3-triol) and fatty acids.
These compounds are significant in biological, pharmaceutical, cosmetic, and industrial applications due to their amphiphilic nature.

CAS Number: Glyceryl monoesters vary; a common example is Glyceryl monostearate: 31566-31-1

Synonyms:
Monoacylglycerol,Glyceryl monoesters,Glyceryl monostearate (for specific compound),Glycerol esters,Fatty acid esters of glycerol,Glycerides

Chemical Structure and Classification
Basic Structure
Glyceryl esters are formed through esterification of one, two, or all three hydroxyl groups of glycerol with fatty acids.

Classification
Monoacylglycerols (MAGs) – One fatty acid attached
Diacylglycerols (DAGs) – Two fatty acids attached
Triacylglycerols (TAGs) – Three fatty acids attached

Chemical Formula
General formula depends on esterification:
MAG: C21H42O4 (e.g., glyceryl monostearate)
DAG and TAGs vary by fatty acid chain length

Physicochemical Properties
Property    Description
Appearance    White to off-white waxy flakes or solid
Odor    Odorless or faint fatty odor
Melting Point    ~55–65°C (for glyceryl monostearate)
Solubility    Insoluble in water; soluble in ethanol and oils
Density    ~0.9 g/cm³
pH (10% dispersion)    ~6.0–8.0
HLB Value (MAGs)    Typically around 3.8–5.0 (lipophilic emulsifier)

Synthesis and Production Methods
Esterification Process
Glycerol is reacted with fatty acids or triglycerides in the presence of an acid or base catalyst.

Transesterification
Triglycerides (vegetable oils) are transesterified with glycerol to yield mono- and di-glycerides.

Enzymatic Methods
Lipase-catalyzed esterification for high selectivity, used in food/pharma industries.

Industrial Raw Materials
Sources: Palm oil, coconut oil, soybean oil, animal fats

Industrial and Commercial Grades
Food Grade

Pharmaceutical Grade
Used as excipients, stabilizers, and bioavailability enhancers

Cosmetic Grade
Emollients and emulsifiers in lotions and creams

Applications
Food Industry
Emulsifiers in bakery, dairy, margarine, and frozen desserts

E471 recognized as safe food additive

Cosmetics and Personal Care
Skin conditioners, opacifiers, and emulsifiers in creams, lotions, and lipsticks

Pharmaceuticals
Bioavailability enhancers

Carrier in suppositories and capsules

Industrial Uses
Lubricants, plasticizers, antistatic agents in plastics

Biological Role and Metabolism
Glyceryl esters are digested into glycerol and free fatty acids by lipase enzymes.
Serve as energy reservoirs in adipose tissues.
Some (like MAGs) have antimicrobial properties.

Analytical Techniques
Gas Chromatography (GC)
Identification and quantification of glyceride species

High-Performance Liquid Chromatography (HPLC)
